I am building a test printing system and I need CMYK output. If I use CMYK in InkScape to fill, the blacks are the addition of CMYK. For example, if I use Pillow and examine a black region,
I get [255, 255, 255, 0]
Is there a way to get black in the print?

You should use Scribus if you want get proper CMYK output.
